5-HTP

5-HTP
The direct serotonin precursor — 200 mg from Griffonia simplicifolia, one step from serotonin.
- Direct serotonin precursor — one step past the tryptophan bottleneck
- Crosses the blood-brain barrier — no transport competition like tryptophan
- 200 mg from Griffonia — the dose used in the human trials
- Mood · sleep · appetite axis — the serotonin substrate, declared
5-HTP is the direct precursor to serotonin — one enzymatic step away, past the rate-limiting bottleneck of L-tryptophan. 200 mg from Griffonia simplicifolia seed.
L-tryptophan has to clear a slow, rate-limiting enzyme before it becomes serotonin — most never makes it. 5-HTP starts past that gate, at a declared 200 mg.
5-HTP converts to serotonin in one enzymatic step — skipping the rate-limiting hydroxylase that caps how much tryptophan ever becomes serotonin.
Serotonin is the precursor to melatonin. An RCT in older adults found 100 mg/day improved sleep-quality components, most in poor sleepers.
For people who want the precursor itself — not a tryptophan supplement competing for transport. Evening-dosed, claim-honest, no overclaiming.
Three peer-reviewed human RCTs — serotonin metabolism, sleep, and the appetite axis.
The active, declared. Calcium carbonate is the carrier; the 5-HTP is the compound.
One step to serotonin. Then the night-time pathway.
5-HTP crosses the gut and the blood-brain barrier, then a decarboxylase converts it to serotonin directly — no rate-limiting hydroxylase in the way.
Serotonin is the substrate for melatonin at night — the pathway behind 5-HTP's evening sleep-quality signal.
The 5-HTP is extracted from Griffonia simplicifolia seed — the natural plant source used across the human trials, at the 200 mg dose.
Serotonin synthesis has its substrate. Some notice an easier evening wind-down and faster sleep onset first.
Mood baseline and sleep quality tend to settle. Evening appetite may ease — the satiety signal from the trials.
The serotonin-substrate effect stabilises — the window where the sleep and satiety RCTs measured their endpoints.
A maintained baseline. Serotonin support holds with continued evening use; the effect reverses on stopping.
Compound-based expectation — not medical advice. Individual response varies.
Two capsules once a day, ideally in the evening 20–30 minutes before a meal with water — the serotonin→melatonin pathway matters most at night. Cumulative; give it several weeks.
Peer-reviewed. Human trials.
12-week RCT (n=30 older adults): 100 mg/day 5-HTP improved sleep-quality components vs control, most prominently in poor sleepers, alongside shifts in gut-microbiota composition.
Sutanto CN et al. Clin Nutr. 2024;43(3):593–602 ↗RCT in overweight women: Griffonia 5-HTP significantly raised 24-hour urinary 5-HIAA (a serotonin metabolite, p<0.001) and increased satiety scores vs placebo over 4 weeks.
Rondanelli M et al. Eat Weight Disord. 2012;17(1):e22–8 ↗Double-blind RCT in obese adults: 5-HTP reduced caloric intake and improved adherence to dietary prescriptions vs placebo — early evidence for the serotonin-appetite axis.
Cangiano C et al. Am J Clin Nutr. 1992;56(5):863–7 ↗Randomised controlled trial: 5-HTP altered serotonergic activity at both neural and behavioural levels on social-cognition tasks — confirming central serotonin engagement in humans.
Zamoscik V et al. Sci Rep. 2021;11:21637 ↗took it for sleep onset. 5-htp because tryptophan competes for transport and most never converts. evening dose, 30 min before food. week 2, falling asleep faster, fewer 3am wake-ups. the sutanto sleep rct sold me. not sedating, just easier wind-down.
5-htp is the precursor one step from serotonin, past the rate-limiting enzyme. griffonia source, 200mg, the trial dose. three weeks in, evening mood is steadier and night-time appetite is easier to control. clean label, declared dose.
works for me on sleep, but do NOT stack with an ssri or maoi — serotonin isn't something to combine blindly. i'm on nothing so fine. 4 stars only because it's gelatin caps, not vegan, which they should fix. the sleep-latency effect is real.
the cangiano and rondanelli data is on satiety — 5-hiaa up, late-night snacking down. week 4, evening cravings noticeably lower. didn't expect that part. mood steadier too. evening dose works best for me.
first week nothing. by week 3 sleep was deeper and i woke less foggy. if you want to knock yourself out this isn't that — it's serotonin substrate working with your own system. taking it nightly. 4 stars: real but gradual.
magnesium glycinate for the gaba side, 5-htp for the serotonin/melatonin side — different mechanisms, no overlap. best sleep consistency i've had in a while. 6 weeks, both staying in the rotation.
